The total synthesis of the transition metal pteridine (TMPT) ligand of the molybdenum and tungsten enzymes would be completed, then metal complexes synthesised for spectroscopic and biological comparisons with the natural systems.The synthesis of structurally modified analogues of the TMPT, including quinoxalines and pteridines, would be followed by formation of transition metal complexes from these, and then their electrochemical, spectroscopic and biological evaluation aimed at gaining information about the following aspects of the mechanism of action of these enzymes:a What geometry at the metal is essential for catalytic oxygen transfer activity?b What is the role of TMPT(s) in the catalytic process?c What is the form of TMPT at the active site during catalytic turnover?d Does the structure of the organic ligand remain the same throughout the catalytic process?e Is there interplay between the oxidation state of the metal and that of the TMPT?
