This proposal is to develop a synthetic route to the marine natural product Spongistatin 1 / Altohyrtin A, a highly potent antitumour agent. One of the spiroketals in the molecule will be formed using methodology developed within the group whereby an anomeric sulphone is alkylated with an epoxide and the resultant enol ether allowed to spirocyclise to the thermodynamically favoured configuration. Dithianes will also be exploited as masked carbonyl anion equivalents, deprotection of which will allow spirocyclisation to occur within the constraining framework of the macrocycle. This tactic should result in the spiroketal adopting the required non-fully anomerically stabilised configuration of the natural product. The synthesis will be flexible allowing formation of the macrocycle either through macrolactonisation or a ring closing metathesis. Flexibility will also allow the order of fragment coupling reactions to be varied thus maximising our options.
