We have recently discovered the unexpectedly selective formation of both tetrahydrofurans and tetrahydropyrans, using cyclisation precursors which are either 1, 2-diols or their derivatives. As such systems are easy to prepare using asymmetric dihydroxylation methods, if these observations prove to be general, then this could constitute an attractive general strategyto these systems. In particular, tetrahydrofuran and tetrahydropyran units can be found in many matural products with potentially useful biological activity. Following the results of methodology investigations, the total synthesis of appropriate target structures will be addressed, with particular emphasis on the synthesis of antifungal natural products and analogues thereof.
