The project will involve the development and application of a new approach to capillary electrochromatography-mass spectrometry (CEC/MS), using on-line laser time-of-flight mass spectrometry (TOFMS). A key feature of our work will be the development of a novel interface to transfer species separated on the CEC column into the vacuum region of the mass spectrometer. This will involve direct laser desorption of eluants at the end of the capillary column, using a pulsed CO2 laser. Desorbed species will be selectively ionised, with high efficiency, using a second pulsed UV laser and the resulting product photons accumulated in an rf quadrupole trap, followed by pulsed extraction and time-of-flight mass analysis. This approach will circumvent problems due to matrix ions from the eluting solvent encountered with electrospray interfaces. In parallel work will be carried out (1) to study the flow and chromatographic characteristics of capillary separation systems using packed columns (porous silica gel) having mean particle diameters in the range 0.5 to 2 micron, (2) to produce capillaries containing reticulated stationary phase supports, and (3) to develop in-situ derivatisation techniques. Our initial applications using this hybrid instrument will focus on ultratrace analysis of chemical carcinogens and pharmaceutical compounds in small volume samples.
