Summary on Grant Application Form
A DAD/HPLC system will be interfaced to an electrospray mass spectrometer, resulting in acquisition of triply coupled data; problems of aligning data, adjusting flow rates and comparing output from both systems will be tackled, resulting in a single software package. Factor analysis will be performed both in MS and EAS directions, and various criteria will be established to improve the methodology, taking into account the need to recover similar elution profiles from both techniques. Extensive simulations will be performed. Real date, consisting of degradation mixtures of chlorophyll will be generated, and used as initial case studies. Systematic experiments will be performed to produce a variety of different degradation mixtures. The influence of resolution, signal to noise, spectral similarity, etc, on the effectiveness of factor analysis will be studied, resulting in improvements in methodology. A second case study will be found. Guide-lines as to how good data quality must be and how this may influence the approach to factor analysis will be established. A small package will be produced.
