One of the major objectives in transition metal-catalysed olefin metathesis research at present is to understand the factors that control the cross metathesis of functionalised alkenes so that conditions and catalysts can be developed which facilitate the use of this reaction in organic synthesis. This objective is worthwhile as success would provide synthetic organic chemists with a strategically new and attractive alkene synthesis. In view of the current high levels of industrial and academic interest in amino acids and peptides, we propose to undertake cross metatheseis studies using unsaturated amino acids as substrates. In order to demonstrate the methodology we develop, we propose to apply it to the synthesis of several biologically active amino acids and to prove its potential for the modification of peptides both in a tradition and combinatorial sense.
