The aim of this research is to develop a novel latent-active glycosylation strategy. This glycosylation strategy is based on isomerisation of substituted allyl glycosides to give vinyl glycosides which can subsequently be user in Lewis acid-mediated glycosylation reactions. This approach will enable the preparation of complex oligosaccharides of biological importance in a highly convergent manner.The work will include: (i) the preparation of a range of allyl glycosides which can readily be isomerised to the corresponding vinyl glycosides and these substrates will be used to synthesise both a- and b-glycosidic linkages; (ii) the use of reactive Rh and Ni catalysts, which will isomerise more efficiently the allyl glycosides and which will allow the isomerisation and glycosylation to be performed as a one pot procedure; (iii) the preparation of allyl glycosides containing a non-chiral allyl substituent such that they can be used in the active-latent glycosylation strategy; (iv) the development of methodology to convert vinyl glycosides into other glycosyl donors; (v) the preparation of oligosaccharide fragments of natural importance using the novel latent-active glycosylation strategy.
