Computational methods now being developed (6) to determine the degree of selective binding, of enantiomers to a silica-based chiral stationary phase will be extended to the design of the active groups for use in (reverse-phase) electrochromatography. The groups will be synthesized and bonded on to small diameter (1-3m) silica particles in cooperation with Phase Separations Ltd who will provide advice (Dr P Myers), raw materials, and some of the special facilities for synthesis.The particles will then by packed (3) into fused silica capillaries with supercritical CO2 as carrier; the density of the packed bed will be controlled by varying the CO2 density. The performance of the resulting columns for the separation of a variety of racemic mixtures of pharmaceutical, crop-protection and food industry interest will be investigated first in HPLC and then electrochromatographic mode, Systemmatic investigations will be made of other factors affecting electrochromatographic performance, eg field strength; buffer concentration and pH:ionic strength; and organic modifier.
