The initial objective is to fully define the scope and limitations of iodocyclisations of N-(3-alkenyl)arylsulphonamides, via an overall 5-endo-trig process, leading to highly substituted pyrrolidines. Both (E)- and (Z)-isomers will be examined and the mechanism of isomerization tof 2,5-trans substituted pyrrolidines to the corresponding 2,5-cis isomers will be similarly defined. This flexible and rapid approach can be readily adapted to asymmetric synthesis and examples of this will be provided in target syntheses aimed at members of the Kainoid group of natural neuroexcitators and the immunostimulant indolizidine alkaloid Cyclizidine. Extensions to the corresponding 3-alkenysulphides will also be investigated with a view to preparing homochiral tetrahydrothiophene derivatives. This section will be exemplified by a new, short approach to (+)-Biotin. If time allows, the preparation of isoxazolidines from related cyclisations of unsaturated hydroxylamine derivatives will also be investigated.
