We propose to use natural metabolites as precursors in the synthesis of biodegradable hypercoiling polymers. The target polymer is one which contains both hydrophobic and either weak acid or weak base pendant groups, polymerised through ester or amide linkages. Such a polymer forms a hypercoiling macromolecule capable of being degraded in vivo into harmless metabolites. By selecting different pendant groups and varying the chain length of hydrophobic side residules it is possible to custom synthesise polymers which uncoil or hypercoil at specific pH values. Synthetic work in these targets areas will be coupled with structural and behavioural (particularly dynamic surface tension) characterisation. By understanding the detailed relationship between structure and behaviour in this way structures will be tailored to meet the specific requirements for clinical applications (Synthetic lung surfactants, polymeric antibacterials, targetted drug delivery).
