Our previous studies of the luciferase system entrapped in agarose gels will be extended to include enzyme 'dots', where the enzyme is immobilised on a small surface area. This will be achieved in collaboration with the Department of Electronics and Electrical Engineering, University of Glasgow, who have experience of the technology. The fundamental enzymic properties of surface-bound luciferase will be evaluated in a single dot device as a prelude to the investigation of possible uses of arrays of such dots. Surface attachment will allow improved spatial resolution for image storage since the 10 mm dots will be individually addressable, eg by optical fibres. Two colour photoactivation of enzyme dots will be achieved by caging both ATP and luciferin such that each responds to a different wavelength of light. This facility will be exploited in studies of pattern recognition using masks. By attachment of luciferase to electrode surfaces we will examine the effects of local electrochemical generation of oxygen and protons as modulators of enzyme activity as well as the effects of voltage and current. variable gain light amplification will be achieved by enzyme switching. Photolysis of glyceraldehyde phosphate dehydrogenase caged with a novel photolytic group will be used to generate ATP.
